Patient-derived organoids (PDOs) and cell line-derived organotypic three-dimensional (3D) cultures better recapitulate key features of tumor architecture. These PDOs and organotypic 3D cultures better mimic the structure, function, microenvironment, and expression profiles of the primary tumor, representing a powerful tool for precision medicine and the discovery of new therapeutic targets. This special issue aims to integrate cutting-edge research across molecular biology, transcriptomics, epigenetics, and non-coding RNAs with a particular emphasis on the discovery of potential therapeutic targets. We welcome original research and reviews on the promises and challenges of PDOs and 3D organotypic cultures and their applications in tumor modeling, drug screening, and personalized medicine in cancer.
